PyroMAT® System, a Suitable Solution for the Detection of a Wide Range of Pyrogens


The PyroMAT® system is a Monocyte Activation Test (MAT) for the detection of pyrogens in parenteral drugs and medical devices. 

Unlike the rabbit pyrogen test and bacterial endotoxin test, also known as the LAL Test, the PyroMAT® system does not require the use of animals. In addition, the PyroMAT® system can detect a broad range of pyrogens, whereas the LAL test only detects endotoxins (a specific type of pyrogen found in Gram-negative bacteria).

The PyroMAT® system uses the Mono-Mac-6 (MM6) cell line as its source of monocytes. This cell line has been shown to express the various toll-like receptors (TLRs) responsible for the detection of pyrogens.

Several TLR ligands were used to assess the sensitivity of PyroMAT® to each of them. Ligands targeting both extracellular and endosomal TLRs were successfully and reproducibly detected with three batches of MM6 cells, confirming the cell line’s ability to detect a wide range of pyrogenic substances.
